Ubrogepant is an orally administered calcitonin gene-related peptide (CGRP) receptor antagonist, commonly known as a "gepant," developed for the acute treatment of migraine with or without aura in adults. It works by selectively blocking CGRP receptors, thereby reducing migraine pain and associated symptoms without causing the vasoconstriction associated with triptan medications. The development of ubrogepant followed growing evidence of the critical role of CGRP in migraine pathophysiology and advances in CGRP-targeted therapies. Originally discovered and developed by Allergan, ubrogepant demonstrated favorable efficacy and safety in clinical trials, leading to its approval by the U.S. Food and Drug Administration (FDA) in December 2019 under the brand name Ubrelvy. Its approval marked the first oral CGRP receptor antagonist available for the acute treatment of migraine, providing an important therapeutic option for patients who do not respond to or cannot use conventional migraine medications.
